Chemiluminescent substrates are important tools in the detection of proteins and nucleic acids. Essentially, chemiluminescence blot detection starts when the primary antibody recognizes and binds to the target protein or nucleic acid immobilized on a membrane. This is followed by a secondary antibody labeled with an enzyme such as horseradish peroxidase (HRP) or alkaline phosphatase (AP). A chemiluminescent substrate for the particular enzyme is then applied to the blot causing a light emitting reaction which is then detected, signaling the presence of the target.
