Develop your own TR-FRET custom assays and label your biomolecule with a Europium lanthanide chelate. LANCEĀ® Europium chelates use the following reactive groups for labeling chemistry:
- Isothiocyanate (ITC)-activated chelates react primarily with free amine groups, including the N-terminus of a protein or peptide as well as lysine residues on antibodies, proteins, and peptides.
- Iodoacetamide (IA)-activated chelates react with free sulfhydryl groups (including cysteines) on peptides and proteins to form a stable, covalent thio-ether bonds.
- Diochlorotriazine (DTA)-activated chelates react with free amino, sulfhydryl, and hydroxyl groups and are suitable for labeling proteins, peptides, nucleic acids, and small molecules containing these functional groups.
LANCE Eu-W1024 chelates are preferred in most High Throughput Screening (HTS) applications due to their low fluorescent background and high S:B in TR-FRET assays. For assays that require high concentrations of EDTA or other chelators, a high temperature, or a pH below 7, the Eu-W8044 chelate offers greater stability.
