AlphaScreen SureFire Bead Mix Volumes
top
The following links contain information on volumes to use when preparing buffer/bead master mixes that are used for running 50-500 wells. The tables list recommended volumes of each component, based on the number of assay points you will be running. Though the ratio of the components will remain the same for a given kit, the total volume of master mix being added to each well depends on whether you are using the adherent cell protocol or the non-adherent cell protocol. The suggested volumes assume that you are either manually pipetting or using a repeat pipette (no troughs, no lab automation).
Master mix recipes for 1-step kits
- Check that the manual for your kit says to "Mix Reaction buffer (60 parts), Activation buffer (10 parts) and Donor and Acceptor beads (1 part each)" in the "Buffer preparation and storage conditions" table.
- Adherent cell protocol
- Non-adherent cell protocol
Master mix recipes for most 2-step kits
- Check that the manual for your kit says to "Mix Reaction buffer (40 parts), Activation buffer (10 parts) and Acceptor beads (1 part)" in the "Buffer preparation and storage conditions" table.
- Adherent cell protocol
- Non-adherent cell protocol
Master mix recipes for some 2-step kits
- Check that the manual for your kit says to "Mix Reaction buffer (45 parts), Activation buffer (5 parts) and Acceptor beads (1 part)" in the "Buffer preparation and storage conditions" table.
