The AlphaLISA® SureFire® Ultra™ total mTOR assay kit can be used to measure levels of mTOR (both phosphorylated and non-phosphorylated) in cellular lysates using no-wash Alpha technology.
This kit has been pre-validated for use in combination with Pelago Bioscience’s CETSA® (Cellular Thermal Shift Assay) technology. Please note that further assay optimization will be required in order to adapt the kit to your particular cell line and experimental conditions. A valid CETSA® licence from Pelago Bioscience or a single kit agreement is required for using the CETSA® technology.

In the AlphaLISA® SureFire® Ultra™ assay, Donor beads are coated with streptavidin to capture one of the antibodies, which is biotinylated. Acceptor beads are coated with a proprietary CaptSure™ agent that immobilizes the other antibody, labeled with a CaptSure™ tag. In the presence of target protein, the two antibodies bring the Donor and Acceptor beads close together, generating signal. The amount of light emission is directly proportional to the amount of protein present in the sample.
