While cell-based assays are increasingly used for kinase screening, biochemical assays remain an advantageous tool in the development of kinase modulators as they provide a method to directly assess the activity of a compound in a pure system. Biochemical assays allow insights into the mechanism of action of candidate compounds that may be important to achieve the desired kinase inhibition characteristics and selectivity.
While one way to identify kinase inhibitors of therapeutic interest is to monitor phosphorylated substrate formation, a quicker and universal approach is to monitor the ATP consumption after the kinase assay reaction. easylite-Kinase™ is a detection system that uses luciferin and a luciferase to produce a luminescent signal in proportion to the amount of ATP remaining after stopping the kinase activity. In the easylite-Kinase system, kinase inhibitors will prevent ATP consumption by the kinase, and the ATP remaining will generate a positive luminescent signal.
Luminescence assays have the advantage of being quick and easy to read without requiring microplate readers with specialized excitation sources or filters. The luminescence signal produced gives a high signal-to-background ratio, which expedites assay development time.
