AequoZen FroZen Cells, kappa opioid receptor (OP2), human recombinant, CHO-K1

Our frozen, irradiated AequoZen cells express a variety of GPCRs which all couple to a calcium response. Upon GPCR stimulation and subsequent calcium binding to the aequorin oxidation of coelenterazine leads to emission of light. No cell culture is required prior to testing, for these frozen cells.

  • Step-by-Step protocols for culture-free functional testing on our validated cells
  • Affordable access to small aliquots of cells (minimum size: 10 million cells/vial)
  • Ready-to-use cells for Profiling, Pharmacology or Screening applications
  • Up-scaling and customized developments

The AequoScreen technology is a generic GPCR technology which can be used with Gs, Gi and Gq coupled GPCRs and calcium coupled ion channels. Following receptor stimulation, increases in intracellular calcium enable measurement of the resulting flash luminescence signal.

The "frozen cells" approach to functional testing consists in the dissociation of cell culture from testing. That means that we will culture the cells and freeze them with an optimized protocol to stop all replication. These cells are then stored in liquid nitrogen or simply in a -80°C freezer, thawed and used directly in a functional, cellular GPCR test with previously validated performance.

Application Note

AequoScreen aequorin-encoding Plasmids for performing aequorin luminescent assay starting from non-aequorin cell lines

Aequorin is a photo protein originating from the jellyfish Aequorea Victoria. The apo-enzyme (apoaequorin) is a 21 kD protein, which requires a hydrophobic prosthetic group, coelenterazine, to be converted to aequorin, the active form of the enzyme. This enzyme possesses 3 calcium binding sites which ...
